Trace Amines and Neurological Disorders: Potential Mechanisms and Risk Factors explores trace amines which, under normal conditions, are present in the mammalian brain and peripheral nervous tissues at very low (nanomolar) concentrations. However, in a diverse array of human pathologies ranging from substance abuse, depression, attention deficit hyperactivity disorder, eating disorders, schizophrenia, and other neurological and neuropsychiatric diseases, the levels of trace amines are unusually high with an imbalance in their functions. Furthermore, the rapid turnover of trace amines is evidenced by their dramatic increases following treatment with monoamine oxidase inhibitors (MAOI) or deletion of the MAO genes. This suggests that the concentration of trace amines may be considerably higher at neuronal synapses than predicted by steady-state measures, implicating some pathophysiological role. Therefore, understanding molecular mechanisms and developing selective agonists and antagonists for trace amine-associated receptors (TAARs) has become a good approach for treating these diseases. Although the effects of trace amines at low physiological concentrations in mammalian species have been difficult to demonstrate, they may serve to maintain the neuronal activity of other monoamine neurotransmitters by possessing postsynaptic modulatory effects, particularly dopamine and serotonin, within defined physiological limits. Such an effect of trace amines makes them ideal candidates for the development of novel therapeutics for a wide range of human disorders. This book presents up-to-date, cutting-edge, and comprehensive information on the link between trace amines and neurological disorders. Focuses on recent findings on trace amines, providing insights into the functional significance, molecular mechanisms, and biological relevance of TAARS in neurological disorders Edited work with chapters authored by leaders in the field from around the globe, the broadest, most expert coverage available Provides cutting-edge research on trace amines-mediated signaling in vertebrate model systems

Medicinal chemistry is both science and art. The science of medicinal chemistry offers mankind one of its best hopes for improving the quality of life. The art of medicinal chemistry continues to challenge its practitioners with the need for both intuition and experience to discover new drugs. Hence sharing the experience of drug research is uniquely beneficial to the field of medicinal chemistry. Drug research requires interdisciplinary team-work at the interface between chemistry, biology and medicine. Therefore, the topic-related series Topics in Medicinal Chemistry covers all relevant aspects of drug research, e.g. pathobiochemistry of diseases, identification and validation of (emerging) drug targets, structural biology, drugability of targets, drug design approaches, chemogenomics, synthetic chemistry including combinatorial methods, bioorganic chemistry, natural compounds, high-throughput screening, pharmacological in vitro and in vivo investigations, drug-receptor interactions on the molecular level, structure-activity relationships, drug absorption, distribution, metabolism, elimination, toxicology and pharmacogenomics. In general, special volumes are edited by well known guest editors.

Drug Discovery Approaches for the Treatment of Neurodegenerative Disorders: Alzheimer’s Disease examines the drug discovery process for neurodegenerative diseases by focusing specifically on Alzheimer’s Disease and illustrating the paradigm necessary to ensure future research and treatment success. The book explores diagnosis, epidemiology, drug discovery strategies, current therapeutics, and much more to provide a holistic approach to the discovery, development, and treatment of Alzheimer’s Disease. Through its coverage of the latest research in targeted drug design, preclinical studies, and mouse and rat models, the book is a must-have resource for all pharmaceutical scientists, pharmacologists, neuroscientists, and clinical researchers working in this area. It illustrates why these drugs tend to fail at the clinical stage, and examines Alzheimer’s Disease within the overall context of improving the drug discovery process for the treatment of other neurodegenerative disorders. Provides a compilation of chemical considerations required in drug discovery for the treatment of neurodegenerative disorders Examines different classes of compounds currently being used in discovery and development stages Explores in vitro and in vivo models with respect to their ability to translate these models to human conditions Distills the most significant information across multiple areas of Alzheimer’s disease research to provide a single, comprehensive, and balanced resource

How to understand your clients' true illnesses, not just their DSM checklists. Though the DSM discusses the criteria for mood disorders in absolute terms—either present or absent—professionals are aware that while such dichotomies are useful for teaching, they are not always true in practice. Recent genetic data support clinicians' longstanding recognition that a continuum of mood disorders between unipolar and bipolar better matches reality than a yes/no, bipolar-or-not approach. If we acknowledge that continuum, how does this affect our approach to diagnosis and treatment? In A Spectrum Approach to Mood Disorders, nationally recognized expert James Phelps provides an in-depth exploration of the signs, symptoms, and nuanced presentations of the mood disorder spectrum, focusing on the broad gray area between Major Depression and Bipolar I. Combining theoretical understanding and real-world scenarios, Phelps offers practical treatment guidelines for clinicians to better understand the subtle ways mood disorders can show up, and how to find the most beneficial path for treatment based on the patient's individual pattern of symptoms. Is it trauma, or is it bipolar? Borderline? Both? Phelps's expertise and wealth of personal experience provides readers with unparalleled insight into a subject that is by nature challenging to define. His emphasis on non-medication approaches, as well as chapters on all the major pill-based treatments (from fish oil to lithium to the avoidance of atypical antipsychotics and antidepressants), creates a comprehensive resource for any clinician working with patients on the mood spectrum. Appendices on the relationship between bipolar diagnosis, politics, and religion; and a plain-English approach to the statistical perils of bipolar screening, offer further value. Phelps has written an invaluable guide of the critical information professionals need to treat patients on the mood disorder spectrum, as well as a useful tool for highly motivated families and patients to better understand the mood disorder that effects their lives. This book seeks to alter the black and white language surrounding these mood disorders to influence a shift in how patients are diagnosed—to insure that treatment matches their specific needs.

Diet and exercise have long been recognized as important components of a healthy lifestyle, as they have a great impact on improving cardiovascular and cerebrovascular functions, lowering the risk of metabolic disorders, and contributing to healthy aging. As a greater proportion of the world’s population is living longer, there has been increased interest in understanding the role of nutrition and exercise in long-term neurological health and cognitive function. Diet and Exercise in Cognitive Function and Neurological Diseases discusses the role and impact that nutrition and activity have on cognitive function and neurological health. The book is divided into two sections. The first section focuses on diet and its impact on neurobiological processes. Chapters focus on the impacts of specific diets, such as the Mediterranean, ketogenic and vegan diets, as well as the role of specific nutrients, fats, fatty acids, and calorie restriction on neurological health and cognitive function. The second section of the book focuses on exercise, and its role in maintaining cognitive function, reducing neuroinflammatory responses, regulating adult neurogenesis, and healthy brain aging. Other chapters look at the impact of exercise in the management of specific neurological disorders such Multiple Sclerosis and Parkinson’s Disease. Diet and Exercise in Cognitive Function and Neurological Diseases is a timely reference on the neurobiological interplay between diet and exercise on long-term brain health and cognitive function.
