This foundational work comprehensively examines the current state of the genetics, genomics and brain circuitry of psychiatric and neurological disorders. It consolidates discoveries of specific genes and genomic regions associated with these conditions, the genetic and anatomic architecture of these syndromes, and addresses how recent advances in genomics are leading to a reappraisal of the biology underlying clinical neuroscience. In doing so, it critically examines the promise and limitations of these discoveries toward treatment, and to the interdisciplinary nature of understanding brain and behavior. Coverage includes new discoveries regarding autism, epilepsy, intellectual disability, dementias, movement disorders, language impairment, disorders of attention, schizophrenia, and bipolar disorder. Genomics, Circuits, and Pathways in Clinical Neuropsychiatry focuses on key concepts, challenges, findings, and methods in genetics, genomics, molecular pathways, brain circuitry, and related neurobiology of neurologic and psychiatric disorders. Provides interdisciplinary appeal in psychiatry, neurology, neuroscience, and genetics Identifies key concepts, methods, and findings Includes coverage of multiple disorders from autism to schizophrenia Reviews specific genes associated with disorders Discusses the genetic architecture of these syndromes Explains how recent findings are influencing the understanding of biology Clarifies the promise of these findings for future treatment

Developing Therapeutics for Alzheimer's Disease: Progress and Challenges provides a thorough overview of the latest advances toward the development of therapeutics for Alzheimer’s disease, along with the major hurdles that still must be overcome and potential solutions to these problems. Despite the lack of progress toward developing therapeutics that can slow or stop the progression of this disease, important discoveries have been made and many promising approaches are advancing in preclinical studies and clinical trials. This book outlines the special challenges related to specific targets and approaches, while presenting a realistic, comprehensive and balanced view of drug discovery and development in this area. Written by international leaders in the field, the book assesses prospects for the emergence of effective agents and allows readers to better understand the challenges, failures, and future potential for research in Alzheimer’s disease. This book is a valuable resource to academic scientists carrying out translational research in Alzheimer’s disease, industrial scientists engaged in Alzheimer's drug discovery, executives in biopharmaceutical companies making strategic decisions regarding the direction of internal research and potential outside partnerships, and graduate-level students pursuing courses on Alzheimer's therapeutics. Provides a realistic but promising assessment of the potential of various therapeutic approaches to Alzheimer’s disease Focuses primarily on neuroprotective agents and cognitive enhancers, as well as approaches to targeting the amyloid B-peptide, tau and Apolipoprotein E Discusses alternative approaches, preclinical and clinical development issues, related biomarkers and diagnostics, and prevention and nonpharmacological approaches

Neurodevelopmental disorders (NDDs) are severe, heritable conditions characterized by impaired brain development and disability in cognition, social interaction and behavior. Genetic predisposition appears to interact with environmental factors to produce the onset of the disease. In the last decade genomic analysis on NDDs advanced the knowledge of the genetic causes of these disorders: most of the genes identified were not predicted from known biology, and almost all of them engage in neurodevelopmental processes. However, the same studies revealed an unexpected outcome regarding the molecular etiology: many of the same susceptibility genes as well as molecular pathways are emerging across hitherto unrelated disorders- such as autism and schizophrenia- challenging how we conceptualize these conditions. These new discoveries call for the establishment of additional criteria for the classification of the genes and related phenotypes. Beyond the classical distinction according to diagnostic criteria, new parameters should be taken into account: the genetics, the neurobiology of the gene products and the molecular mechanisms that they control. Our broad understanding of NDDs, requires a combination of approaches, including human studies, animal models and theoretical computation. Indeed, progress in technology and the generation of accurate models are advancing the field, and the research is rapidly growing. This Research Topic presents recent advances in neurodevelopmental disorders from genes to systems, in single conditions and across different phenotypes, and report the newest treatments in use. Recognized experts in the field discuss recent findings in molecular studies, in vivo imaging, neuropsychology, neuropharmacology and computational neuroscience. This multidisciplinary approach enriches our understanding of the mechanisms involved in the onset and progression of NDDs with the perspective of revealing the biological signs of the disorders and identifying new therapeutics.

In this up-to-date survey and critical assessment of transgenic and knockout models in neuropsychiatry and behavior, a panel of leading researchers comprehensively assesses how and whether the genetic abnormalities produced from these models manifest the neuropsychiatric disorders to which they correspond. The authors focus on transgenic and knockout models of neurocognitive dysfunction and neuropsychiatric dysfunction. The discussion of neurobiological problems covers mental retardation, polyglutamate, and speech disorders, as well as disorders that involve cognitive, social, speech, and language dysfunction. The neuropsychiatric dysfunctions examined include psychosis and schizophrenia, anxiety, depression, and bipolar disorder.

The discipline of neurology emerged in the second half of the 19th Century. With it, chairs and departments of neurology, training programs, specialized journals, and new societies came into being. Trying to understand disorders of the nervous system, however, has roots that can be traced back into antiquity, and the new discipline did not develop in similar ways throughout the world. Further, whereas some neurological disorders seemed relatively easy to understand even before there was a neurological examination, others posed challenges, and many still remain shrouded in mystery. The authors of the present volume examine the fascinating prehistory of neurology, its emergence with as an independent discipline, and how it developed throughout the world. They also look at a number of neurological disorders, some sensory, others motor, and still others affecting higher cognitive functions, to illustrate how our understanding of neurological disorders has changed over time. With 55 chapters, many covering material that has received little or no coverage in other books, this history of neurology is unique in its breadth and depth. Filling a great void, its pages are laced with fascinating medical facts, information about people, and cultural connections. This volume is sure to appeal to neurologists, historians of science and medicine, and inquisitive people from other fields --- readers who wish to understand the roots of a discipline and the challenges faced by its pioneers.

Discovered little more than a decade ago, optogenetics - a revolutionary technique combining genetic and optical methods to observe and control the function of neurons - is now a widely used research tool. Optogenetics-driven research has led to insights into Parkinson's disease and other neurological and psychiatric disorders. With contributions from leaders and innovators from both academia and industry, this volume explores the discovery and application of optogenetics, from the basic science to its potential clinical use. Chapters cover a range of optogenetics applications, including for brain circuits, plasticity, memory, learning, sleep, vision and neurodegenerative and neuropsychiatric diseases. Providing authoritative coverage of the huge potential that optogenetics research carries, this is an ideal resource for researchers and graduate students, as well as for those working in the biotechnology and pharmaceutical industries and in a clinical setting.
