Over the last few years several new instrumental techniques have been introduced for chromatographie separations. In addition, rapid developments in existing methods, such as preparative HPLC, have taken place. Despite these advances, how ever, a handbook covering the various preparative aspects ofthe new separation tech niques does not exist. This book is an attempt to fill the gap and to present a compila tion of modern separation techniques that will be useful for researchers faced with day-to-day preparative problems. Numerous examples of separations have been selected in order to show the possibilities (and also the limits) of each technique treated. These are often either applications from our own laboratory or else they reflect the approach we have been following for the isolation of natural products from plant sourees. Owing to the large number of published papers and the diversity of secondary plant constituents. an exhaustive survey of the literature has not been undertaken. We hope, however, that the examples selected will suggest to the reader which technique(s) and which conditions to choose for a particular isolation problem in the field of natural products. For invaluable help in the preparation of the manuscript for this book, we would like to thank Corinne Appolonia and Christine Marston. Lausanne, November 1985 K. Hostettmann M. Hostettmann A . . Marston Foreword Although not many people realize this, isolation and purification of biologically active materials is becoming increasingly crucial.

Completely revised and substantially extended to reflect the developments in this fast-changing field. It retains the interdisciplinary approach that elegantly combines the chemistry and engineering involved to describe the conception and improvement of chromatographic processes. It also covers recent advances in preparative chromatographic processes for the separation of "smaller" molecules using standard laboratory equipment as well as the detailed conception of industrial chemical plants. The increase in biopharmaceutical substances is reflected by new and revised chapters on different modifications of continuous chromatography as well as ion-exchange chromatography and other separation principles widely used in biochromatography. Following an introductory section on the history of chromatography, the current state of research and the design of chromatographic processes, the book goes on to define the general terminology. There then follow sections on stationary phases, selection of chromatographic systems and process concepts. A completely new chapter deals with engineering and operation of chromatographic equipment. Final chapters on modeling and determination of model parameters as well as model based design, optimization and control of preparative chromatographic processes allow for optimal selection of chromatographic processes. Essential for chemists and chemical engineers in the chemical, pharmaceutical, and food industries.

Chiral Chromatography Thomas E. Beesley Advanced Separation Technologies Inc., Whippany, New Jersey, USA Raymond P. W. Scott Chemistry Department, Georgetown University, Washington DC, USA and Chemistry Department, Birkbeck College, University of London, UK Analytical techniques based on separation processes, such as chromatography and electrophoresis, are finding a growing range of applications in chemical, pharmaceutical and clinical laboratories. The Wiley Separation Science Series provides the analyst in these laboratories with well-focused books covering individual techniques, so that they can be applied more efficiently and effectively to contemporary analytical problems. The different enantiomers of a drug can exhibit widely different physiological activity in degree and nature. As a result, the separation and identification of enantiomers is now a very important analytical problem and chiral chromatography is the natural technique to apply to the resolution of such mixtures. Chiral Chromatography provides the reader with a basic understanding of the nature of chromatographic separations and relates the principles specifically to the separation of enantiomers. The following information is included: * chiral separations involving both gas and liquid chromatography * descriptions of the apparatus used for both techniques * detailed discussion on the retention mechanism that results in chiral selectivity * the structure and synthesis of a wide range of chirally active stationary phases used in both gas and liquid chromatography * preparative applications for large scale purification of enantiomers * applications of capillary electrophoresis and capillary electrochromatography. In addition to the above, a large number of examples of the separation of both commercially and physiologically interesting chiral mixtures are given, as is a detailed discussion on the mechanism of selectivity of each example. Thomas Beesley was founder and is the CEO for a leading manufacturer of chiral stationary phases and has published papers on TLC, HPLC and chiral separations involving cyclodextrins. He has also coauthored papers with Daniel W. Armstrong, an expert on modern cyclodextrin columns. Raymond Scott has worked in the field of separation science for over 40 years and has contributed extensively to the development of both gas and liquid chromatography publishing over 160 papers on the subjects.

Principles and Practice of Modern Chromatographic Methods, Second Edition takes a comprehensive, unified approach in its presentation of chromatographic techniques. Like the first edition, the book provides a scientifically rigid, but easy-to-follow presentation of chromatography concepts that begins with the purpose and intent of chromatographic theory - the “what and why that are left out of other books attempting to cover these principles. This fully revised second edition brings the content up-to-date, covering recent developments in several new sections and an additional chapter on composite methods. New topics include sample profiling, sample preparation, sustainable green chemistry, 2D chromatography, miniaturization/nano-LC, HILIC, and more. Contains thorough chapters that begin with an updated schematic overview and a visual representation of the content Avoids the obfuscation of different terminologies and classification systems that are prevalent in the area, such as the relationship between liquid chromatography and column chromatography Provides integrated and comprehensive topic coverage based on chromatographic bibliometrics and survey reports on the relative usage of chromatographic techniques

Explains why modern supercritical fluid chromatography (SFC) is the leading "green" analytical and purification separations technology. Modern supercritical fluid chromatography (SFC) is the leading method used to analyze and purify chiral and achiral chemical compounds, many of which are pharmaceuticals, pharmaceutical candidates, and natural products including cannabis-related compounds. This book covers current SFC instrumentation as it relates to greater robustness, better reproducibility, and increased analytical sensitivity. Modern Supercritical Fluid Chromatography: Carbon Dioxide Containing Mobile Phases covers the history, instrumentation, method development and applications of SFC. The authors provided readers with an overview of analytical and preparative SFC equipment, stationary phases, and mobile phase choices. Topics covered include: Milestones of Supercritical Fluid Chromatography; Physical Properties of Supercritical Fluids; Instrumentation for SFC; Detection in SFC; Achiral SFC Method Development; Chiral SFC Method Development; and Preparative Scale SFC. The book also includes highlights of modern applications of SFC in the final chapters—namely pharmaceuticals, consumer products, foods, polymers, petroleum-related mixtures, and cannabis—and discusses the future of SFC. Provides a clear explanation of the physical and chemical properties of supercritical fluids, which gives the reader a better understanding of the basis for improved performance in SFC compared to HPLC and GC Describes the advantages of SFC as a green alternative to HPLC and GC for the analysis of both polar, water-soluble, and non-polar analytes Details both achiral and chiral SFC method development, including modifiers, additives, the impact of temperature and pressure, and stationary phase choices Details why SFC is the premier modern preparative chromatographic technique used to purify components of mixtures for subsequent uses, both from performance and economic perspectives Covers numerous detectors, with an emphasis on SFC-MS, SFC-UV, and SFC-ELSD (evaporative light scattering detection) Describes the application of SFC to numerous high-value application areas Modern Supercritical Fluid Chromatography: Carbon Dioxide Containing Mobile Phases will be of great interest to professionals, students, and professors involved in analytical, bioanalytical, separations science, medicinal, petroleum, and environmental chemistries. It will also appeal to pharmaceutical scientists, natural-product scientists, food and consumer-products scientists, chemical engineers, and managers in these areas.
